NLRP3 inflammasome in sepsis (Review)

نویسندگان

چکیده

Sepsis is an imbalanced response to infection that leads life-threatening organ dysfunction. Although increasing number of anti-inflammatory drugs are available, the options for treating sepsis remain limited. Therefore, it imperative understand pathogenesis and pathophysiology develop novel therapeutic targets treat this state. The Nod-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome a cytoplasmic high-molecular weight protein complex composed sensor NLRP3, adapter apoptosis-related speck-like pro-caspase-1. It functions by cleaving pro-caspase-1 become active caspase-1, resulting in maturation release IL-1β IL-18. Activation NLRP3 necessary innate immune defense also serves important role adaptive responses. Studies have shown involved occurrence evolution other inflammatory diseases. present paper reviews activation pathways biological function sepsis, with aim provide basis further research.

NLRP3 inflammasome and its inhibitors: a review

Inflammasomes are newly recognized, vital players in innate immunity. The best characterized is the NLRP3 inflammasome, so-called because the NLRP3 protein in the complex belongs to the family of nucleotide-binding and oligomerization domain-like receptors (NLRs) and is also known as "pyrin domain-containing protein 3". The NLRP3 inflammasome is associated with onset and progression of various ...

The NLRP3 inflammasome and stroke.

Inflammasome pattern recognition receptors, which belong to the family of multi-meric proteins, play an important role in innate immunity, including NLRPs, NLRC, and NAIP. Among these receptors, NLRP3 (nucleotide-binding domain (NOD)-like receptor protein 3) inflammasome may activate the inflammation and participate in atherosclerosis, pathophysiology of myocardial infarction, resultin ischemia...

NLRP3 Inflammasome and MS/EAE

Inflammasomes are cytosolic sensors that detect pathogens and danger signals in the innate immune system. The NLRP3 inflammasome is currently the most fully characterized inflammasome and is known to detect a wide array of microbes and endogenous damage-associated molecules. Possible involvement of the NLRP3 inflammasome (or inflammasomes) in the development of multiple sclerosis (MS) was sugge...

Exogenous Carbon Monoxide Decreases Sepsis-Induced Acute Kidney Injury and Inhibits NLRP3 Inflammasome Activation in Rats

Carbon monoxide (CO) has shown various physiological effects including anti-inflammatory activity in several diseases, whereas the therapeutic efficacy of CO on sepsis-induced acute kidney injury (AKI) has not been reported as of yet. The purpose of the present study was to explore the effects of exogenous CO on sepsis-induced AKI and nucleotide-binding domain-like receptor protein 3 (NLRP3) in...

Cardiolipin and the Nlrp3 inflammasome.

The Nlrp3 inflammasome is a key controller of the proinflammatory cytokine IL-1β. Iyer and colleagues (2013) demonstrate that Nlrp3 activators promote an interaction between cardiolipin in mitochondria and Nlrp3. Cardiolipin is thus a hydrophobic danger signal that, upon translocation to the outer mitochondrial membrane, activates Nlrp3, promoting inflammation.
